General description

Fibroblast growth factor-basic (FGF-Basic) is found in basement membranes and sub-endothelial extracellular matrix. FGF-Basic is induced early in the development of the embryo where it is necessary (but not sufficient) for pluripotency and self-renewal of embryonic stem cells (ESC) and lineage defined stem cells; wherein it functions to support proliferation and inhibit differentiation. Self-renewal of hESC depends upon activation of the Activin/Nodal/Smad2,3 branch and suppression of the BMP/GDF/Smad5 branch of the TGFβ-family cell signaling program. FGF-Basic is a competence factor for Nodal support of pluripotency. Activin-A is necessary and sufficient to maintain stemness. It induces the expression of both Nodal and FGF-Basic in human ES cells.

Application

Fibroblast growth factor-basic (FGF-Basic) is required for the maintenance of human embryonic stem cells in culture, and various other stem cell lines, such as mesenchymal stem (stroma) cells (MSC).[1][2][3]

Physical form

Lyophilized from 0.2 μm filtered solution in 20 mM Tris and 1 M NaCl, pH 7.0, containing 50 μg bovine serum albumin per 1 μg BFGF.

Analysis Note

The bioactivity of FGF-Basic was measured in a fluorometric assay using the redox sensitive dye, Resazurin.

  1. What is the volume of the vial and how do I reconstitute the powder?

    1. The volume of the container is 2 mL. Reconstitution of this product should be performed using 0.2 µm filtered 20 mM Tris, pH 7.0, to prepare a stock solution of >= 25 ug/mL. This stock solution may be diluted immediately before use to achieve the final working concentration. Additional filtration is not recommended, as it may result in product loss due to adsorption onto the filter membrane.

  2. Hey! I am using this product to prepare a stem cell media. I know the concentration is 0.025mg but it has two data sheets, one that its soluble in water (0.250ml) and other that says to dissolve in 20mM Tris Base pH 7.0. How many volume should I add?

    1. The solubility of this product is tested in water. It is lyophilized from a solution containing 20 mM Tris and 1M NaCl with 50 ug BSA as a stabilizer. The product data sheet recommends reconstituting in the 20 mM Tris for optimum stability and long term storage.
