General description

ゼラチンは親水コロイドです。ウシまたはブタの骨と皮が市販品の原料です。ゼラチンはコラーゲン誘導体であり、主成分としてのタンパク質と無機質および水で構成されています。

Application

ブタ皮膚由来のゼラチンは以下の用途に使用されています：
  • A375細胞の培養およびゼラチン分解アッセイにおけるガラス底プレートのコーティングのため
  • 脳切片を用いる組織学的研究におけるガラススライドのコーティングのため
  • 3D印刷されたヒドロゲル上に細胞を封入するためのゼラチンメタクリン酸（GelMA）プレポリマーの合成に
  • 3Dプロジェクションバイオプリンティングのためのチオール官能化ゼラチン（GelSH）ヒドロゲルを調製するため

ブタ皮膚由来のゼラチンは、1～5 μg/cm2または0.5～50 μg/mLで、細胞培養基層として使用することが推奨されます。至適濃度は、細胞のタイプならびに用途と研究対象によって大きく異なります。

Biochem/physiol Actions

ゼラチンポリマーは、細胞や組織の培養に用いられるコラーゲンに代わるバイオマテリアルです。増粘剤および食品成分として有用な、高度に選別されたゲル化剤です。ゼラチンは、発泡剤および結合剤としても用いられ、医薬品業界でも活用されています。生分解性かつ生体適合性に優れ、特に心臓や血管の組織再生用バイオマテリアルにおいて生物医学的用途があります。

Preparation Note

この製品はブタの皮膚に由来します。ゼラチンは冷水よりも熱湯に溶けます。 アルコール、クロロホルム、二硫化炭素、四塩化炭素、エーテル、ベンゼン、アセトン、およびオイルなどほとんどの有機溶媒にはほとんど溶けません。 Bloom（ブルーム）ゲロメーターにより測定されたBloom数は、既知の濃度の溶液から形成されたゲルの強度の指標です。 Bloom数は平均分子量に比例します。 豚表皮ゼラチンのBloom数は90～300 gの範囲で異なります。この製品のゲル強度は300であり、50 mg/mLで水に溶けます。

ゼラチンは、コラーゲン中に存在する平均分子量が大きい水溶性タンパク質の不均一な混合物です。 水中で適切な皮膚、腱、靭帯、骨などを煮沸することでタンパク質が抽出されます。 A型ゼラチンは酸硬化組織由来です。 B型ゼラチンは石灰硬化組織由来です。

Disclaimer

乾燥ゼラチンは、室温で気密容器に保存した場合、長年変わらないままです。 空気の存在下で100℃に加熱すると、膨潤して柔らかくなり、ピリジン塩基およびアンモニアの発生を伴い崩壊して炭素質の塊になります。
