General description

αα-微管蛋白，又称微管蛋白α 4a（TUBA4A），位于人类染色体2q35。该基因编码含有448个氨基酸的α-微管蛋白家族成员。TUBA4A属于微管蛋白亚家族，微管蛋白是微管的主要组成部分。α-亚基的分子量为50,000。

抗乙酰化微管蛋白（小鼠IgG2b同种型）单克隆抗体源自杂交瘤6-11B-1，而后者是由小鼠骨髓瘤细胞与紫海胆(Strongylocentrotus purpuratus)外臂来源乙酰微管蛋白免疫小鼠的脾脏细胞融合产生的。微管蛋白是一种异二聚体，由α-微管蛋白和β-微管蛋白组成；这两个亚基的分子量均为50000，且同源性很高。

Biochem/physiol Actions

乙酰化微管蛋白与多种生物体中的轴丝组装相关。乙酰化微管蛋白可能在微管结构和功能的分化中具有关键作用。α-微管蛋白的翻译后修饰，即通过特定的乙酰化酶向Lys40可逆地添加乙酰基，产生乙酰化微管蛋白。

该抗体还可用于检测原生动物、植物、无脊椎动物和脊椎动物等多种生物体的乙酰化α-微管蛋白。该抗体可识别氨基酸乙酰化后Lys⁴⁰四个残基内衣藻轴突α-微管蛋白α3亚型上的表位。用该抗体在鸡、果蝇和酵母的绝大多数α-微管蛋白中检测到的序列与衣藻中检测到的非常相似，但在某些α-微管蛋白亚型中，相应区域明显不同。偶然情况下，抗体识别的表位可能缺失或被掩盖，就像在大鼠袋鼠上皮样细胞系PtK₂中一样。
