Descrição geral

A shell of 24 protein subunits (apoferritin) and a core Fe 3+ ions with a peptide MW of 440 kDa. Review of the ferritin-like superfamily of iron-storage proteins, virtually ubiquitous in life on earth.

Aplicação

Ferritin from equine spleen has been used:
  • in mycobacterial growth assay[1]
  • in size-exclusion chromatography[2][3]
  • in labeling of macrophage cells[4]
  • as a reference standard in muon spin spectroscopy[5]

Ações bioquímicas/fisiológicas

A ubiquitous iron storage protein that plays a key role in iron metabolism. It serves as an intracellular iron reserve (particularly in spleen, liver, intestinal mucosa, and bone marrow) and functions in iron detoxification.
A ubiquitous iron storage protein that plays a key role in iron metabolism. It serves as an intracellular iron reserve (particularly in spleen, liver, intestinal mucosa, and bone marrow) and functions in iron detoxification. Studies have shown that ferritin iron incorporation is mediated by a ferroxidase activity associated with ferritin H subunits and a nucleation center associated with ferritin L subunits. Release of iron from ferritin has an essential role in iron-dependent lipid peroxidation and may contribute to free radical-induced cell damage in vivo. Therefore, by binding iron, ferritin may function as an antioxidant.

Ferritin is a globular protein that consists of 24 subunits forming a hollow nanocage structure that naturally stores iron oxyhydroxides. Elimination of iron atoms to obtain the empty protein called apoferritin is the first step to use this organic shell

Iron and oxygen chemistry is mediated by iron proteins for many biological functions. Carboxylate-bridged diiron enzymes including ferritin have the common mechanism of oxygen activation via peroxodiferric intermediates. Questions

1–8 of 8 Questions
  1. Hi, I've just purchased a Ferritin from equine spleen (liquid, 100mg, cas No:9007-73-2, MDL No: MFCD00081593), however, I didn't find how many mL this solution is on the product label. Can you please provide any information regarding volume? Thanks.

    1 answer
    1. The concentration, and thus the volume, of the Ferritin will vary for individual lots. The concentration can be found on the lot specific Certificate of Analysis, which can be retrieved by entering the lot number here:
      https://www.sigmaaldrich.com/product/sigma/f4503#product-documentation

The volume can be determined using the following formula:
Units provided / Units per mL = mL provided

The concentration of the Ferritin is currently specified to be ≤125mg/ml

  2. Does it run as a 44kDa specie or 21kDa+19kDa in size exclusion chromatography?

    1 answer
    1. This product is not analyzed by chromatography. However, historical data indicates the material runs primarily at 20-22kD and 13-15kD in SDS electrophoresis studies. Additional bands appear at the 3000 - 5000 molecular weight range.

      Helpful?

  3. What is the concentration of salt(s) in the saline solution carrying the protein?

    1 answer
    1. The saline solution is a 0.15 M sodium chloride solution. The concentration of 0.15 M can also be written as 8.8 mg NaCl per mL, or the textbook "0.9%" for normal saline.

      Helpful?

  4. Hi, I was wondering if I could get the volume of ferritin present in each of the package sizes? Thank you

    1 answer
    1. The concentration of this product is lot specific and will impact the final volume received. Typical lots range from 50 - 75 mg/mL. Please see the link below to review the product specification sheet:
      https://www.sigmaaldrich.com/specification-sheets/115/477/F4503-BULK________SIGMA____.pdf

See below for potential volume ranges for each package size:
      F4503-25MG (0.5 - 0.33 mL)
      F4503-100MG (2 - 1.33 mL)
      F4503-500MG (10 - 6.66 mL)
      F4503-1G (20 - 13.33 mL)
