Beschreibung

Allgemeine Beschreibung

Interferon gamma is a pro-inflammatory cytokine produced by activated T lymphocytes and natural killer cells. It is important in the activation of macrophages and MHC expression and plays a key role in both inflammation and autoimmune disease by enhancing antigen presentation, increasing leukocyte trafficking and affecting cell proliferation and apoptosis.

The SMC^® Human IFN-γ High Sensitivity Immunoassay Kit contains all reagents required to perform the assay. This assay takes advantage of a standard immunoassay workflow configured in a standard plate format.
Assay protocols are similar to existing sandwich ELISA methods with two key differences:
1) Elution buffer disrupts the sandwich, separating the labeled detection antibody for quantification.
2) The Erenna and SMCxPRO^® immunoassay systems detect analytes using Single Molecule Counting (SMC^®) technology.

Anwendung

The sensitivity of the Single Molecule Counting (SMC^® technology gives the SMC^® Human IFN-γ High Sensitivity Immunoassay Kit unparalleled performance in the quantification of IFN-γ in human serum and plasma.

This new streamline kit has been improved with the following features over the legacy IFN-γ kit, #03-0049-00:

• LLOQ has been lowered from 0.2 pg/mL in the legacy kit to 0.03 pg/mL in the new kit
• Improved options for sample prep and incubation
• Lyophilization of the standard provides the kit at one storage condition
• Removal of a plate transfer step
• Increased reagent volumes to accommodate automation
• Validated to run on both SMCxPRO^® and Erenna
• Tested against WHO standard

Note: Recombinant protein is not stable and vortexing is not recommended.

Species Reactivity:
Although IFN-γ was detectable in rat, mouse, non-human primate, feline, and canine plasma, the assay has not been validated for these species.

Cross Reactivity:
The SMC^® Human IFN-γ High Sensitivity Immunoasay Kit was not found to be cross-reactive with the following:

• rhIFN-α2
• rhIFN-β
• rhIFN-λ1 (IL-29)
• rhIFN-λ2 (IL-28A)
• rhIFN-λ3 (IL-28B)
• rhIFN-ω2

Accuracy:
Minimum of 5 serum samples spiked with average recovery of 104% with a range of 92-124%
Minimum of 5 plasma samples spiked with average recovery of 98% with a range of 87-110%
