Allgemeine Beschreibung

Nuclear receptor subfamily 4 group A member 2 (UniProt: P43354; also known as Immediate-early response protein NOT, Orphan nuclear receptor NURR1, Transcriptionally-inducible nuclear receptor) and is encoded by the NR4A2 (also known as NOT, NURR1, TINUR) gene (Gene ID: 4929) in human. Nurr-1 belongs to the nuclear hormone receptor family that acts as a transcriptional regulator and is considered to be important for the differentiation and maintenance of meso-diencephalic dopaminergic (mdDA) neurons during development. It is also crucial for expression of a set of genes such as SLC6A3, SLC18A2, TH and DRD2 which are essential for development of mdDA neurons. It is expressed in a number of T cell, B cell and fibroblast cell lines, but its expression is higher in brain tissue. Two isoforms of Nurr-1 have been reported that are produced by alternative splicing. The ligand-binding domain (LBD) of Nurr-1 contains no cavity as a result of the tight packing of side chains from several bulky hydrophobic residues in the region normally occupied by ligands. It also lacks a classical binding site for co-activators. However, it has hydrophobic region on the surface of LBD, which is critical for its activity. (Ref.: Volakakis N et al (2006). J. Mol.
