Preparation Note
2段階グルタルアルデヒド法 (Avrameas, S., et. al. Scand.J. Immunol, 8, Suppl. 7, 7 (1978))により調製しました。
-20℃で長期間保存します。連続使用する場合、本製品は2～8℃で最大1ヵ月間保存できます。長期間保存する場合、溶液をワーキングアリコートにして-20℃で凍結させることができます。凍結融解の繰り返し、または「自動霜取装置付き」冷凍庫での保存はお勧めしません。長期間の保存で軽微な濁りが生じた場合は、使用前に遠心分離により溶液を澄明化します。
Application
ヒト乳房手術検体から得た表皮タンパク質抽出物が、TBSt/5%ミルクで1：80000に希釈した二次抗体としてのHRP抱合ウサギ抗マウスIgGを用いてウエスタンブロットに供されました。
抗マウスIgG（全分子） - ペルオキシダーゼ抗体は、ウエスタンブロッティング、免疫組織化学的検査およびタンパク質ピンアレイアッセイに使用されています。
抗マウスIgG（全分子）–ペルオキシダーゼ抗体は、1：40,000のワーキング抗体希釈で直接ELISAに使用される場合があります。免疫ブロッティングでは、1：80,000～1：160,000の希釈を用いる場合があります。ホルマリン固定パラフィン包埋ヒト扁桃腺またはヒト虫垂切片の免疫組織化学的検査には、1：200のワーキング抗体希釈を推奨します。マウス筋肉組織切片の免疫組織化学的検査のために、マウスIgGペルオキシダーゼ抗体が1：20の希釈で使用され、同じ研究で1：500の希釈がサウス-ウエスタンブロッティングに用いられました。ヒト骨格筋組織抽出物の免疫ブロッティングでは、1：5000の希釈が用いられました。
Biochem/physiol Actions
IgG抗体は、細菌、真菌およびウイルスが引き起こす感染を防御します。母体のIgGは、感染に対する新生児の免疫防御に不可欠な胎盤を通じて胎児に移されます。IgG抗体は、補体系活性化においてIgM抗体と同様の機能を有します。
