The Serum Triglyceride Determination Kit (Catalog Number TR0100) offers a comprehensive solution for assessing glycerol, true triglycerides, or total triglycerides in serum or plasma samples. I would like to measure the concentration of free glycerol in the culture supernatant of adipocytes using this reagent with a plate reader instead of a cuvette. What are recommended sample and reagent volumes for a plate reader? Have you ever tested?

This product has not been tested for use with a well plate format. The product instructions call for a total volume of 830 µL, where 10 µL each of water, glycerol standard, and sample are added to cuvettes containing 800 µL of Free Glycerol Reagent. Most 96-well plates have a working volume of 100-300 µL, so the volume of each component would need to be scaled accordingly. Adding 3 µL each of water, glycerol standard, and sample to wells containing 240 µL of Free Glycerol Reagent would result in the same concentrations with a total volume of 249 µL. Regardless, the functionality of a plate reader in this application cannot be guaranteed and would need to be assessed independently.

I have an old version of this product (6428) - just one bottle not a kit. I need to reconstitute this but can't find instructions. Can you let me know what to reconstitute in and how much? Thank you!

There are no specific instructions for 6428. F6428 is one bottle of reagent as well. The user guide of F6428 may serve as a reference. A typical reconstitution procedure of F6428 is: Reconstitute the Free Glycerol Reagent with 40 ml of water. After addition of water, stopper the vials, and immediately mix several times by inversion. DO NOT SHAKE. Protect the reagent from light by storing in amber bottles. Please check page 2 of the user guide here:

How is shipping temperature determined? And how is it related to the product storage temperature?

Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.
