General description
ヒトIgGは、2本の同じ軽鎖と一対の同一重鎖を含む糖タンパク質抗体です。 IgGにはIgG1～IgG4の4つの異なるアイソフォームがあります。これらの抗体は、アレルギーや病原体感染に対する免疫応答を調節します。また、IgGは補体固定や自己免疫疾患との関与も示唆されています。
抗体を抗ヒトIgG抗血清から分離するには、ヒトIgGのFab画分に特異的に結合しない免疫グロブリンなどを含むヤギ血清タンパク質を本質的にすべて除去するために、免疫特異的精製を行います。抗ヒトIgGをペルオキシダーゼに抱合させた後、さらに精製して非抱合物質を除去します。

Application
ヤギで産生された抗ヒトIgG（Fab特異的）-ペルオキシダーゼ抗体は、次の用途で使用されています。
抗ヒトIgG抗体は、PBSで1：10,000に希釈されて使用され、室温で1時間インキュベートされました。ELISAアッセイでの結合は、O-フェニレンジアミン基質溶液（Sigma）を用いて検出されました。
- ウエスタンブロット法
- Fabフラグメントを産生するように遺伝子操作された酵母培養物の分析を目的とした、希釈率1：40,000における直接酵素結合免疫吸着測定法（ELISA）。
- 免疫組織化学
- ドットブロット法
- 免疫組織化学（ホルマリン固定、パラフィン包埋切片、1:200）
