General description
A solução salina tamponada de Dulbecco (DPBS) é uma solução salina balanceada (BSS) usada para o manuseio e a cultura de células de mamíferos. O tampão DPBS é usado para irrigar, lavar e diluir células de mamíferos enquanto mantém o equilíbrio osmótico. Ele fornece água e íons inorgânicos essenciais para o metabolismo celular normal. DPBS com cálcio e magnésio facilitam a ligação e a aglomeração celular. A PBS de Dulbecco sem esses íons pode ser usada para lavar e enxaguar células suspensas. Tamponação com fosfato mantém o pH no intervalo fisiológico.
Application
A solução salina tamponada de Dulbecco tem sido usada:
- para diluição de eritrócitos frescos para estudar o aprisionamento de agregados de eritrócitos.
- para lavar os poços de uma placa de microtitulação.[1]
- para lavar as células antes de ressuspendê-las no tampão de lise para isolamento de DNA.[2]
- como um componente do meio de armazenamento de embriões para a vitrificação de embriões.

This product is a sterile-filtered M Quality Level 300 product. It is not molecular biology grade and does not have specifications such as RNase and DNase free. Therefore, this product cannot be considered RNase free for use in in situ hybridization.

What is a Salt Toxicity Test?

The Cell Culture assay is called a Salt Toxicity Test (previously called Cell Culture Salt Testing). A more complete description is below:Cells are harvested and counted. They are resuspended in DPBS and incubated at 37°C, 5% CO2 for 1 hour. The cells are counted and %viability determined. The cells are then cultured for an additional 7 hours (total of 8 hours). The cells are counted again and the % viability determined. The viability at 1 hr has to be >95% of the original viability. The viability at 8 hrs must be >75% of the original viability.
