General description

Laminins are non-collagenous adhesive glycoproteins with a substantial molecular weight. They are heterotrimeric proteins with three disulfide-linked polypeptides known as the α, β, and γ chains. The multiple polypeptide domains of laminins can bind to multiple cell surface receptors, including several integrin dimers, dystroglycan and syndecans, and other ECM components. This extracellular matrix multidomain trimeric glycoprotein is the main component of basal lamina that supports adhesion, proliferation and differentiation.

Application

Laminin from Engelbreth-Holm-Swarm murine sarcoma basement membrane has been used in:
  • insulin-producing cell (IPC) differentiation.
  • Infection inhibition assays.
  • Proliferation Assay.
Laminin supports growth and differentiation of many cell types including epithelial, endothelial, neural, muscle and liver cells. It is recommended for use as a cell culture substratum at 1-2 μg/cm2. The optimal concentration does depend on cell type as well as the application or research objectives.

Biochem/physiol Actions

Laminin proteins are integral components of structural scaffolding in animal tissues. They associate with type IV collagen via entactin and perlecan and bind to cell membranes through integrin receptors, dystroglycan glycoprotein complexes and Lutheran blood group glycoproteins. Laminin has active domains for collagen binding, cell adhesion, heparin binding, and neurite outgrowth fragment.

Preparation Note

This product is supplied at a concentration of 1 − 2 mg/mL in 50 mM Tris-HCl, with 150 mM NaCl. It is filtered using a 0.2μm filter. Thaw this solution slowly before use at 2-8°C to avoid gel formation. For use as a coating, dilute in a balanced salt solution, coat culture surface with a minimal volume and incubate at 37°C for two hours. Wash 3 times with PBS before plating cells. Laminin coatings can be stored for one month at 2-8°C.

Laminin is an extracellular matrix multidomain trimeric glycoprotein, and is the main non-collagenous component of basal lamina that supports adhesion, proliferation and differentiation. Laminin is composed of both A, B1 and B2 chains, which are connected by many disulfide bonds. This laminin product was isolated from mouse Engelbreth-Holm-Swarm tumor.

  1. Can the plates to which I add laminin be stored for a certain amount of time—for example, a week in the refrigerator? The thing is, when I make them fresh, I sometimes miscalculate how many slices I need to use, and I always end up wasting some. Thankss

    1. Sterile laminin-coated plates/coverslips may be stored at 2–8°C for approximately 1 month. Store coated items sealed to avoid excess drying. Do not use if discoloration or spider-web formations are observed on the coated surface. Please see the link below to review additional information available in the product datasheet:

  2. How long can stocks be frozen at -20C for?

    1. In undiluted form or diluted to working concentration, this material may be stored at -20°C in aliquots up to the recommended retest date for the specific lot. This data is published in the Certificate of Analysis. Avoid repeated freeze/thaw cycles. The solution may be transferred to smaller vials in working volumes for long-term storage.     1. The product L2020 will be shipped using dry ice. Upon receipt, it should be stored at -20 degrees Celsius.

  4. How is this laminin concentration determined? (by which method?) I tried to measure the concentration of this product by ELISA, and the results always came out half of what is written on the vial.. what is the protein purity?

  5. Can I coat Laminin, Product L2020, on top of poly-L-lysine for better adherence?

    1. When coating coverslips or slides for use with neural stem cells (neurospheres), the following procedure is recommended. Precoat the coverslips with poly-L-lysine (Product No. P5899) at 100 μg/mL. This is incubated for 30 minutes or more at room temperature. The coverslips are then washed with sterile water and allowed to dry. The poly-lysine coated coverslips can be stored at room temperature until ready to use (up to ~1 month). Coverslips are then coated with 30 - 50 μL of warmed laminin, which is at a concentration of 50 μg/mL. Incubate at 37°C for 30 minutes. Remove the laminin, and wash with DME (50 μL) twice. Cells are then added to the coverslips.

  6. How can I store solutions of Laminin, Product L2020?

    1. The product has a recommended storage temperature of -20°C.  Please note, the product needs to be thawed slowly at 2-8°C.  If the product has been warmed up at room temperature and it gels, it cannot be reactivated for use. We recommend storage at 2-8°C for up to one week. For longer storage, the product needs to be frozen at -20°C in single-use aliquots.

  8. What type of laminin is Product L2020?

    1. In the nomenclature system by Burgeson, this is Laminin 1. The laminins were numbered with Arabic numerals in the order discovered. The previous A, B1, and B2 chains, and their isoforms are listed by the Greek letters, for alpha, beta, and gamma, respectively, followed by an Arabic numeral to identify the isoforms.  For example, the first laminin identified from the Engelbreth-Holm-Swarm tumor (EHS) was designated laminin-1 with the chain composition α-1/β-1/γ-1. The genes for these 3 chains are LAMA1, LAMB1, and LAMC1. Reference: Burgeson, R.E., et al., A new nomenclature for the laminins. Matrix Biol., 14, 209-211 (1994).
