General description

α−Mannosidase from Canavalia ensiformis is a tretamer composed of two subunits that each contain two components at 44 and 66 kDa.
Lysosomal α-mannosidases are class II mannosidases that belong to glycoside hydrolase family 38 and play an important role in the degradation of asparagine-linked carbohydrates of glycoproteins.
α-Mannosidase is an exo-glycosidase and an inc metalloenzyme.

Application

α-Mannosidase can be used to liberate mannose from a variety of synthetic and natural α-mannosides. It has also been used in a study to investigate the causes of neurodegeneration in mucolipidosis II ′knock-in′ mice.
Liberates mannose from a variety of synthetic and natural α-mannosides.
α-Mannosidase from Canavalia ensiformis (Jack bean) has been used:
  • for the digestion of neutral glycans.
  • to remove O-glycans from recombinant β-glucosidase
  • for the inhibition of renal fibroblasts proliferation

Biochem/physiol Actions

α−Mannosidase has been shown to inhibit the proliferation of B-lymphocytes.
α-Mannosidase is an acid hydrolase which is located in plant vacuoles and is thought to be involved with the turnover of N-linked glycoproteins.
α-Mannosidase cleaves near α1−2-, α1−3- and α1−6-linked mannose residues.

Physical form

Suspension in 3.0 M (NH4)2SO4 and 0.1 mM zinc acetate, pH 7.5

Other Notes

One unit will hydrolyze 1.0 μmole of p-nitrophenyl α-D-mannoside to p-nitrophenol and D-mannose per min at pH 4.5 at 25°C.

Questions

  1. α-マンノシダーゼ Canavalia ensiformis(タチナタマメ)由来 1mg を以前購入しましたが、何マイクロリットルの何の溶液に溶解しるか教えてください。

    1 answer
    1. This product is a 1 – 10 mg/mL suspension in 3.0 M (NH4)2SO4 containing 0.1 mM zinc acetate. The volume varies from lot to lot and is reported in the product Certificate of Analysis. For example, a lot reporting a concentration of 5 mg protein/ml will have a volume of 200 uL for the 1 mg package size. Please see the link below to review a sample of lot specific Certificate:
