Application

Furosemide has been used:
  • to investigate the influence of timing of dexamethasone administration on auditory hair cell survival in mice[1]
  • to study its effect on intracranial pressure (ICP) after subcutaneous administration in rats[2]
  • as a stimulant for renin release and subsequent increase in plasma angiotensin II (ANG II)[3]

Biochem/physiol Actions

"High ceiling" diuretic that strongly affects renal tubular action by increasing renal blood flow; antihypertensive.
Furosemide can block the Na+/K+/2Cl- co-transporter in the ascending thick loop of Henle. It can enhance the synthesis of intrarenal prostaglandin.[4] It enhances its ototoxicity in animals when used along with kanamycin.[5] Furosemide is linked with thiamine insufficiency in individuals with heart failure.[6]
Inhibits ion co-transport in the kidney.
