No in-house testing has been performed to determine if Calcein is membrane permeable or membrane impermeable. The literature suggests Calcein appears to be membrane impermeable. Conn's Biological Stains, 10th Edition, page 250 states, "entry of the dye into cells may be achieved by direct insertion of the hydrophilic calcein, or by the uptake of calcein AM with intracellular formation of calcein."
