Various solvents, including ethanol, are reported to inhibit forskolin-mediated activation of adenylate cyclase. Dimethyl sulfoxide (DMSO) is the recommended solvent; at a final concentration of 5% or less, little if any inhibition of adenylate cyclase activation will occur. A 50 mM stock solution of forskolin can be prepared by solubilizing 10 mg of forskolin in 488 µL of fresh, high purity, and anhydrous DMSO. After reconstitution, aliquot and freeze at -20°C. The 50 mM stock solution can be diluted 1:100 in DMSO to achieve a 500 µM working stock solution.

Forskolin is soluble in water (with 2% ethanol) up to 0.2 mM by first dissolving in ethanol at 5 mg/mL and performing subsequent dilutions in water. Similar results were obtained with DMSO.
