Biochem/physiol Actions

Cell-permeable diterpenoid that possesses anti-hypertensive, positive inotropic, and adenylyl cyclase activating properties. Many of its biological effects are due to its activation of adenylyl cyclase and the resulting increase in intracellular cAMP concentration. Forskolin effects calcium currents and inhibits MAP kinase.

General description

Forskolin is a cell permeable diterpenoid isolated from Coleus forskohlii.

Questions

1–6 of 6 Questions
  1. How can i prepare 500uM Forskolin (10mg) and where can the prepared solution be stored?

    1 answer
    1. Various solvents, including ethanol, are reported to inhibit forskolin-mediated activation of adenylate cyclase. Dimethyl sulfoxide (DMSO) is the recommended solvent; at a final concentration of 5% or less, little if any inhibition of adenylate cyclase activation will occur. A 50 mM stock solution of forskolin can be prepared by solubilizing 10 mg of forskolin in 488 µL of fresh, high purity, and anhydrous DMSO. After reconstitution, aliquot and freeze at -20°C. The 50 mM stock solution can be diluted 1:100 in DMSO to achieve a 500 µM working stock solution.

  2. What is the shelf life of the solid form of forskolin?

    1 answer
    1. This product is not assigned an expiration date or recommended retest date. Products with no expiration date or recommended retest date should be routinely inspected by customers to ensure they perform as expected. These products are also subject to a one year warranty from the date of shipment. For more information you may access the "Product Dating Information" document under "ADDITIONAL USEFUL DOCUMENTS ABOUT OUR PRODUCTS" at the bottom of the Quality Services page with this link: https://www.sigmaaldrich.com/life-science/quality-and-regulatory-management/quality-services.

  3. How should I store a DMSO or ethanol solution of Product F3917, Forskolin?

    1 answer
    1. Forskolin solutions at 5 mg/mL in either ethanol or DMSO have been found to be stable for at least 6 months at room temperature, as determined by HPLC analysis.

      Helpful?

  4. Will residual solvents inhibit forskolin activation of adenylate cyclase?

    1 answer
    1. Various solvents, including ethanol, inhibit the forskolin activation of adenylate cyclase. DMSO is recommended for preparing forskolin solutions, because at concentrations of 5% or less of DMSO, there is little if any inhibition of forskolin activation.

      Helpful?

  5. How can I make an aqueous solution of Product F3917, Forskolin?

    1 answer
    1. Forskolin is soluble in water (with 2% ethanol) up to 0.2 mM by first dissolving in ethanol at 5 mg/mL and performing subsequent dilutions in water. Similar results were obtained with DMSO.
